Summary: | Objective: To explore the effects of sprouty4 (SPRY4) protein on keratinocyte proliferation and differentiation. Methods: Human immortalized keratinocytes (HaCaT) cells were transfected with SPRY4 protein siRNA, and HaCaT cells without transfection served as controls. Real-time fluorescence quantitative PCR (RT-qPCR) was used to assess expression levels of mRNA for HaCaT cells differentiation (Involucrin, CK1 and CK10), and the CCK-8 assay was used to measure HaCaT cell proliferation. Results: RT-qPCR results showed 94.75% reduction in expression levels of SPRY4 mRNA in comparison to the controls. Moreover, knockdown of SPRY4 decreased expression levels of involucrin, CK1 and CK10 mRNA, while stimulating HaCaT cells proliferation. Conclusion: SPRY4 protein stimulates keratinocyte proliferation, while inhibiting differentiation.
