The effect of arachidonic acid elicitor on phytoalexin stimulation in eggplant callus suspension culture

The aim of this study was to investigate the relationship between the resistance of Verticillium wilt and the amount of phytoalexin, which is the defense molecules synthesized in plant tissues. In this study, Long Purple eggplant variety (disease susceptible) and S. sisymbriifolium wild type (diseas...

description The aim of this study was to investigate the relationship between the resistance of Verticillium wilt and the amount of phytoalexin, which is the defense molecules synthesized in plant tissues. In this study, Long Purple eggplant variety (disease susceptible) and S. sisymbriifolium wild type (disease resistant) were used as plant material. As elicitor, arachidonic acid (5 and 10 mM) was included in the study. Stimulation with elicitor lasted 24, 48 and 72 hours. Extracted substances were analyzed in gas liquid chromatography (GLC). The formation of phytoalexin was not adequately stimulated in the callus suspension culture of the resistant species. The results showed that arachidonic acid was effective in stimulating the formation of phytoalexins in the callus suspension culture in Long Purple variety, and in the in vitro conditions solavetivone was formed in the tissue phase and lubimin substances in the filtrate phase. The highest material accumulations were obtained in both phases: ’10 mM arachidonic acid x 72 hours’ ‘elicitor dose x stimulation period’ interaction was found to be significant.
