description | Objective To explore the effect of LncRNA H19 on endometrial decidualization. Methods The expression of LncRNA H19 in decidual tissues of spontaneous abortion and induced abortion was detected by RT-qPCR; The expression of LncRNA H19 in non-pregnant endometrium and decidual tissues of different gestational weeks was detected; Using MPA and 8-Br-cAMP inducers to induce the transformed human endometrial stromal cells (THESCs), RT-qPCR was used to detect the expression of prolactin (PRL) and insulin-like growth factor binding protein 1 (IGFBP-1); The expression changes of LncRNA H19 and FOXO1 were detected after the process of decidualization; Lentivirus was used to overexpress and interfere with LncRNA H19 in THESCs, and the proliferation of THESCs was detected by CCK-8 assay. After successful lentivirus transfection, decidualization was induced. RT-qPCR was used to detect the expression of PRL, IGFBP-1 and FOXO1 in blank control group, LV-GFP induction group, LV-H19-RNAi induction group and LV-H19 induction group. Results The expression of LncRNA H19 in spontaneous abortion group was lower than that in induced abortion group (P < 0.01); LncRNA H19 reached its peak at the 7th week of decidual tissue in early pregnancy (P < 0.01); After the decidualization of THESCs, the levels of PRL, IGFBP-1, FOXO1 and LncRNA H19 were higher than those of the control group (P < 0.01).The results of CCK-8 assay showed that the proliferation of THESCs was inhibited after the interference with LncRNA H19 (P < 0.01), while the proliferation of THESCs was promoted after the overexpression of LncRNA H19 (P < 0.01). The decidualization induction was performed, after interfering with and overexpressing the LncRNA H19 of THESCs. Compared with the blank control group, the LV-GFP induction group showed the increased mRNA expression of PRL (P < 0.05), IGFBP-1 (P < 0.01) and FOXO1 (P < 0.01). Compared with LV-GFP induction group, the mRNA expression of PRL (P < 0.05) and IGFBP-1 (P < 0.01) in LV-H19 induction group was increased, while the mRNA expression of PRL (P < 0.05), IGFBP-1 (P < 0.01) and FOXO1 (P < 0.01) in LV-H19-RNAi induction group was decreased. Conclusion LncRNA H19 promotes the proliferation and decidualization of endometrial stromal cells.
