description | Introduction: In clinical microbiology laboratories, urine specimens are the most frequently analysed specimens. Rapid and reliable methods are needed especially for no growth culture. In this study, the performance of the Sysmex UF-1000i instrument was compared with the culture results for the purpose of fast and reliable evaluation of the urine specimens.
Materials and Methods: 9525 urine samples sent for urine culture to the Central Laboratory between February 2018-April 2018 were evaluated retrospectively. The culture results and the leukocyte and bacterial results obtained from the fluorescence flow cytometer were matched and statistical analysis was performed by applying SPSS 15.00. ROC curves were plotted to determine the cut-off in 100% negative predictive value and 100% sensitivity to be used for leukocyte and bacterial screening.
Results: 9.6% of the urine specimens were reported to have shown growth, 33.4% were contaminated, and 57% did not show growth. The cut off value with 100% negative predictive value and the highest elimination rate in male and pediatric patients in this study were as follows: bacteria < 50/μL, leukocytes 15/μL, and in female patients as < 50/μL of bacteria and 10/μL. It was determined that 63.63% of the cases in male patients, 52.54% in pediatric patients and 19.06% in female patients did not require a culture study.
Conclusion: The flow cytometry system allows pre-culture screening to be accomplished in significant quantities without being subjected to culture. It will help prevent resistance development by avoiding the use of unnecessary antibiotics. |
