Summary:The performance of the commercially available media Dulbecco's modified Eagles' medium (DMEM} and Roswell Park Memorial Institute medium (RPMI — i 640) in long term and short term primary cell cultures of cattle were compared by culturing foetal kidney fibroblast and lymphocytes. The number of population doublings in fibroblast cultures and mitotic drive in lymphocyte cultures were taken as indicators of efficacy of the media used. It was found that DMEM and RPMI — 1640 were significantly better (P < 0.0I) for cattle fibroblast and lymphocyte cultures respectively
