description Glioblastoma is the most common primary malignant brain tumor in adults. With current standard therapy which includes extensive microsurgical resection along with concurrent chemoradiotherapy and adjuvant temozolomide (TMZ), the median survival of glioblastoma patients is only 14.60 months nowadays. Recent studies demonstrated that human adipose tissue derived mesenchymal stem cells (hAT⁃ MSCs) possessed the glioma ⁃ trophic migratory capacity. The engineered hAT ⁃ MSCs expressing herpes simplex virus⁃thymidine kinase (HSV⁃tk), yeast cytosine deaminase::uracil phosphoribosyltransferase (CDy:: UPRT), and rabbit carboxylesterase (rCE) could exert inhibitory effects on glioma when combined with prodrugs, such as ganciclovir (GCV), 5⁃fluorocytosine (5⁃FC) and irinotecan (CPT⁃11), respectively. hAT⁃ MSCs carrying the oncolytic virus or expressing tumor necrosis factor ⁃ related apoptosis ⁃ inducing ligand (TRAIL) also could inhibit the growth of glioma. This paper summarizes the recent progress in this field to pave the way for hAT⁃MSCs based targeted therapy of glioma in future. DOI:10.3969/j.issn.1672⁃6731.2012.06.005
