description ObjectiveTo investigate the regulatory effect and mechanism of lncRNA XIST on salivary adenoid cystic carcinoma (SACC) cell growth and cisplatin (DDP) chemosensitivity. Methods Human SACC cell lines SACC-83, adenoid cystic carcinoma-2 (ACC-2) and normal submandibular gland cells HSG were cultured to establish DDP-resistant SACC-83/DDP cells. The lncRNA XIST interfering plasmid and miR-429 mimic/inhibitor were respectively transfected into SACC-83 and SACC-83/DDP cells with Lipofectamine 2000. The mRNA expression levels of lncRNA XIST and miRNA(miR)-429 in cells were detected by RT-qPCR. CCK-8, clone formation assay, TUNEL assay and western blot were used to detect the effects of lncRNA XIST and miR-429 on the proliferation, apoptosis and DDP sensitivity of SACC cells, respectively. The targeted relationship between lncRNA XIST and miR-429 was verified by dual-luciferase assay. Results The lncRNA XIST was highly expressed in SACC cells. Interfering lncRNA XIST inhibited the proliferation and promoted the apoptosis of SACC-83 and SACC-83/DDP cells (all P < 0.001). The expression of miR-429 was down-regulated in SACC cells, and lncRNA XIST targeted and negatively regulated the expression of miR-429. Down-regulation of miR-429 reversed the inhibitory effect of shRNA-XIST on the proliferation of SACC cells and its resistance to DDP (all P < 0.05). Conclusion Interfering lncRNA XIST can inhibit the growth of SACC cells and improve the sensitivity of DDP by up-regulating miR-429.
