Summary: | By this article, we are going to report results of a study focused on investigation of interactions of cardiolipin (CL) with cytochrome c (CyT) based on chemometric analyses. Electrochemical and spectroscopic data were recorded and then, they were analyzed by the use of conventional and chemometric techniques. Multivariate curve resolution-alternating least squares (MCR-ALS) was used for simultaneous resolution of different types of electrochemical and spectroscopic data which were augmented into a single data matrix. The outputs of MCR-ALS were used to justify interactions of CL with CyT which completed the results of conventional analyses. Finally, according to the interactions of CL with CyT, two electroanalytical methods including exploiting second-order advantage from differential pulse voltammetric data and an amperometric method were developed for determination of the CyT and according to their performance, the best method was chosen for the analysis of real samples. The DPV responses of the biosensor were linearly correlated with CyT concentration in two ranges of 0–1.5 ng/mL and 1.5–8 ng/mL.
