First, screen keygene module related to the staging of bladder cancer. Then, taking the hubgenes in the key gene module as features, a stage prediction model isconstructed by logistic regression and stochastic gradient descent method. Itshows that themolecular combination of CTHRC1, COL6A3, TIMP2 andSULF1 can be used as an indicator for stage diagnosis. Importantly, the COL1A1is not only the pivot gene in the weighted gene co-expression network, but alsothe core of the protein-protein interaction network. And a set of single-cellRNA sequencing data in Gene Expression Omnibus (GEO) database is used to verifythe results. The consistent conclusions indicate that COL1A1 is a potentialoncogene and biomarker in the progression of bladder cancer, and can be used asa molecular target for the treatment of bladder cancer to slow the rate oftumor development. In addition, function and pathway enrichment analysis showthat focal adhesion is a crucial pathway in the tumor metastasis of bladdercancer, and members of the fibrous collagen family play an important role inthe proliferation and spread of tumor cells. The biomarkers and pathwaysidentified in this study provide an important basis for the determination ofpersonalized medical programs.
