Pharmacodynamics and mechanism of Huyu ointment on the mouse model of allergic contact dermatitis

description | Objective: To explore the effect and anti-inflammatory mechanism of Huyu ointment on the mouse model of allergic contact dermatitis. Methods: 2, 4-Dinitrofluorobenzene was used to sensitize the abdomen of C57 mice. The ears and back of mice were stimulated to establish acute and chronic ACD models. Mice were randomly divided into the blank control group, the model group, Huyu ointment group, the positive group and the matrix group. The histopathological changes of mouse ears were observed. Acute ACD model was used to measure ear thickness and quality of left and right ears. Chronic ACD model was used to detect back skin barrier function and spleen index. PCR measured inflammatory factors TNF-α, IL-1β, IL-6, IL-4, IL-17 mRNA expression levels in skin. Results: In the acute and chronic ACD model, HE staining showed that the Huyu ointment group could reduce the degree of epidermal thickening, reduce edema, and relieve dermal vascular congestion. The toluidine blue staining showed that the Huyu ointment group could reduce the number of mast cells. In the acute ACD model, the ear swelling degree was 0.041±0.010, the mass difference between the left and right ear was 0.845±0.296, while the ear swelling in the model group was 0.156±0.012, the mass difference between the left and right ear was 1.902±0.417, the two groups had a significant difference (t=16.46, 4.62, respectively, both P<0.01). In the chronic ACD model, the SCH of Huyu Ointment group was significantly higher than that of model group (56.99±2.19 vs 18.43±1.74, t=7.78, P<0.05), while the TEWL significantly decreased (17.63±1.59 vs 24.61±1.58, t=-3.12, P<0.05), the spleen index also significantly decreased (0.0031±0.0005 vs 0.0040±0.0005, t=-3.18, P<0.05). PCR results showed that Huyu ointment group can reduce mRNA expression level of TNF-α, IL-1β, IL-6, IL-4 and IL-17 (t=3.39, 2.16, 15.53, 20.24, 2.82, respectively, all P<0.05). Conclusions: Huyu ointment has a good effect on acute and chronic ACD mouse model. Its mechanism may be related to repairing the skin barrier, controling Th1, Th2 immune response, reducing inflammatory factors. |
