description Background: Acorus calamus Linn., an amphibious and amphibiotic plant used in Ayurveda, is also known as vach, sweet flag, and other names. Purpose: The present study aims to developed inclusion complex of essential oil with 2‑hydroxy propyl beta cyclodextrin and evaluate the anxiolytic activity through a light dark box model and elevated plus maze model. Material and methods: The inclusion complex is made with cyclodextrin using a variety of techniques, such as kneading, even though the essential oil has wide spectrum action. Acorus calamus oil was first extracted from freshly gathered leaves using Clevenger equipment for eight hours. The kneading method made inclusion complex, which was then dried in a desiccator for 24 h. Results: This study effort confirms anxiety studies in part due to the chemical constituent of the Acorus calamus known as asarone through GCMS, among other things. TGA, SEM, and XRD analyses are used for confirmation investigations, as well as for confirming the combination of oil with cyclodextrin. The main purpose of cyclodextrin is to improve the stability and bioavailability of the oil. This research confirms that after in vivo studies on Swiss albino mice, cyclodextrin complex with essential oil has anti-anxiety properties with better stability than a Diazepam (Standard Dose) at 1.0 mg/kg by p.o. route. Conclusion: These findings confirm the stability of 2‑hydroxy propyl beta cyclodextrin with essential oil which is valuable candidate for development of alternative anxiolytics with a lower incidence of adverse effect.
