Summary:This study was conducted to determine flaxseed secoisolariciresinol diglucoside (SDG) and its metabolite (enterolactone, EL) effects on DNA methyltransferase (DNMT), histone deacetylase (HDAC), and methyl-CpG-binding domain protein (MBD) expression in reactive oxygen species treated murine adipocytes. SDG or EL treated adipocytes (1 × 107) were treated with hydroxyl or superoxide radical, and then total RNA was extracted and purified using an RNeasy Mini kit. DNMTs, HDACs, and MBD2 expression by real-time quantitative PCR was measured. The fold change in expression of the DNMTs, HDACs, and MBD2 was calculated using 2-ΔΔCT method. The results showed that flaxseed SDG and enterolactone downregulate DNMTs, HDACs, and MBD2 expression. How DNMTs, HDACs, and MBD2 expression was down-regulated by flaxseed SDG and enterolactone is unknown and needs further study.
