Monosaccharide composition analysis of polysaccharides in safflower by ion chromatography-pulsed amperometric detection(离子色谱-脉冲安培法分析西红花多糖的单糖组成)

采用水提醇沉法提取西红花球茎、柱头、花瓣、侧芽等部位的粗多糖,离心干燥得到的水溶性粗多糖在沸水浴条件下经硫酸水解后,通过离子色谱-脉冲安培法测定西红花不同部位多糖的单糖组成.该方法简单高效,灵敏度高,选择性好,不需要气相色谱和毛细管电泳所需的衍生化操作.在优化条件下,测得西红花多糖中含有岩藻糖、鼠李糖、阿拉伯糖、甘露糖、葡萄糖、半乳糖和果糖7种单糖.采用此方法,各单糖检测的线性范围在0.02~20 mg • L-1, 线性相关系数均大于0.999 2,加标回收率在87.2%~111.6%,相对标准偏差在1.26%~3.61%,检出限在3.70~11.35 µg • L-1....
