Summary:In order to improve the germination of somatic embryos of Swietenia macrophylla the effect of the dehydration and the increase of sucrose concentration in culture medium were evaluated. Three times of dehydration (48, 72 and 96 hours) and three sucrose concentration (4.0, 6.0 and 8.0%) were evaluated in independent experiments. After 60 days of culture the number of somatic embryos that remained alive, germinated and showed secondary embryogenesis was quantified. It was verified that the germination of the somatic embryos was affected with the dehydration process; nevertheless, the culture of the embryos during 30 days in culture medium with 4.0 and 6.0% of sucrose and later to transfer them to germination medium increased the number of germinated embryos without affecting the survival and with normal development of bud and root.
