Summary:A single column chromatography separation of a binary system has been investigated and analyzed. The chiral separation process involved separating Guaifenesin enantiomers using Chiralcel OD as the chiral stationary phase. The desired dimensions of the column were investigated independently through simulations and the final proposed dimensions were chosen from commercially available column packed with Chiralcel OD. The simulation was done using MatLab and verified with results obtained from reference. The chromatography process was described using Equilibrium dispersive mathematical model and solved numerically using high accuracy Weighted Essentially Non-Oscillatory (WENO) on finite volume scheme. Flow parameters such as volumetric flow rate etc, were also evaluated and a comparison between two methods of determining cycle time were made.
