description Root-knot nematodes Meloidogyne spp. were the injurious pests in tropical countries. Safe and ecofriendly control tacties is needed to be done. The purposes of this research were: obtaining actinomycetes isolate with enzymmatic activity of damaging the eggshell of root-knot nematodes, uncovering enzymes role in the process of the destruction of a root-knot nematode egg shell, and utilizing isolate actinomysetes was elected as biological control agent to root-knot nematode. Research were done by isolation and selection actinomysetes which capable damaging of root-knot nematode egg, purification and characterization of enzymes (fractional ammonium sulphate and DEAE cellulose) continued testing of the application in the the green house. About 84 isolat actinomysetes shown to have activity chitinolytic , proteolytic and chitino-proteolytic. Based on the results of the test in vitro the best isolates were PSJ-27 (chitinolitic), TL-8 (proteolytic) and TL-10 (chitino-proteolytic). The TL-10 isolate best in preventing immature egg developt, causing premature hatching on mature egg, as well as causing damage to the eggshells and the death of the larvae stage-2. Isolation and purification of enzymes produced by TL-10 isolate are chitinase (43.5 kDa) and protease (24 and 22.5 kDa). Combination treatment of purified chitinase and protease was able to hydrolyzed of egg shells effectively and causing most larvae stage-2 dead (75-100%). On the green house experiment the TL-10 isolate shown the best performance in the pressing root-knot nematode population and inhibits the formation of galls. An increase in the amount of inoculum (density of spores) led to an increase in the effectiveness of the emphasis of root-knot nematode population and inhibition of the formation of galls.The TL-10 Isolate is the best and potentially for biological control agent to rootknot nematodes Meloidogyne spp.
