The aim of this research was to find out the stability of chitosan-loaded GVT-0 in dissolution medium and the ability to enter into MCF-7 cell line.
Preliminary research carried out by purity test through TLC and melting point. GVT-0 preparation was to improve the delivery in the form of nanoparticles in which chitosan medium chain as polymer and tripolyphosphate as cross-linker. Nanoparticle of chitosan-loaded GVT-0 characterization included morphology, distribution and particle size, zeta potential, and entrapment efficiency. Stability test carried out in HCl 0,1 N and artificial intestinal fluid (AIF), whereas cellular uptake assay done by MCF-7 cell line as the model cell.
Results of this research showed that GVT-0 being used was pure by TLC and melting point which were indicated by one single spot in UV 254 and 366 nm and not more than 2°C rises in melting point, chitosan-loaded GVT-0 nanoparticles could be made by ionotropic gelation. The obtained nanoparticles was spherical in shape. The particle size in the range of 30.0-282.2 nm with the polydispersity index ranged from 1.3-10.5, zeta potential were +5.5 upto +11.8 mV. The entrapment efficiency ranged from 49.73 to 58.99%. In vitro stability test showed that nanoparticles stability were pH dependent. The nanoparticles relative unstable in both medium 0.1 N HCl and artificial intestinal fluid. In vitro cellular uptake assay showed that nanoparticle chitosan-loaded GVT-0 were able to enter the MCF-7 cell line. Those results above indicated that nanoparticle may be used to deliver GVT-0 effectively. |
