Summary:The effect of deacetylation degree of chitosan on the effectiveness of chitosan beads as solid support immobilized lipase by cross-linking techique has been carried out. The study aims to obtain optimal conditions for immobilization of lipase on chitosan beads. Chitosan beads made from the chitosan powder with various degree of deacetylation by inverse phase method. The study starts from the optimization process of immobilization such as phosphate buffer pH and concentration of glutaraldehyde as a cross-linker. Lipase immobilized on chitosan beads was tested activities, thermal stability, re-uses and parameter kinetics on the hydrolysis reaction of palm oil triglycerides. Phospate buffer with pH 6 is optimal condition to dissolved lipase enzym. DD chitosan that resulted are 80,71%, 87,51% and 91,77%. Optimum concentration of glutaraldehyde optimal is approached in glutaraldehyde 0,5%(v/v) for DD chitosan bead 80,71% with (%) percentage immobilization enzyme 84%
