Summary: | Wakegi onions can not develop flowers. Therefore, no seeds can be obtained. As
a result, wakegi onion had a narrow genetic diversity. This study aimed to found the
optimum concentration for induction of polyploidy by colchicine in the wakegi onion
and got poliploid of wakegi onion plants. The experiment was conducted at the Faculty
of Agriculture, Gadjah Mada University on June 2011 until January 2013. Experimental
design used was factorial Completely Randomized Block Design 2 factors with 4
replications. The first factor was the cultivar of �Lembah Palu�, �Palasa�, and
�Sumenep�. The second factor was the concentrations of colchicine were 0 gL-1, 0.5
gL-1, 1 gL-1, and 1.5 gL-1 with the long incubation period of three days. Observations
were made at ages 2 to 8 weeks after planting in the bottle for the growth and
morphological parameters and the citology.
The results showed that chromosome number 2n = 32 (tetraploid) obtained at
�Palasa� with 0.5 and 1.5 gL-1 colchicine treatment and that in �Sumenep� with 1 gL-1
colchicine treatment. Poliploid plants mentioned above had a larger size of cell, stomata
and epidermis, and stomata density less than untreated plants. Colchicine-induced plant
growth was slower than plants without colchicine treatment.
