Summary:Trichomonosis is a parasitic diasease caused by Trichomonas gallinae that inhabit anterior digestive tract of columbids. Genetic analysis is needed to determine whether isolates differed genetically depending on geographical location using ribosomal DNA (rDNA) sequences. The aim of this research is to identify T. gallinae based on rDNA sequences by DNA sequencing to analyze the phylogenetic relationships. The DNA sample were isolated from esophageal mucus of pigeon in Yogyakarta. Amplification of rDNA by PCR technique used TRI_1F and TRI_1R primers under condition of pre-denaturation 94°C for 3 min, denaturation 94ºC for 30 s, annealing 52ºC for 45 s, extension 72 ºC for 1 min, and final extension 72 ºC for 5 min at 35 cycles. The PCR reaction produced 351 bp DNA segment which has followed by sequencing. Then result of rDNA segment sequences would be treated in multiple alignment with other protozoas taken from Genbank and then analyzed using MEGA5.1 Result of this research showed that the sample has closer relationship with isolate Colorado and Spain A from Genbank indicated by genetic distance of 0- 0,3%.
