because they have good savor for the body. The main obstacle in the dairy goat
livestock is the increase of subclinical mastitis subclinical disease. Subclinical
Mastitis is an internal tissue inflammation of the udder gland without any clinical
symptoms found either milk or udder, but an increase in the number of
inflammatory cells, was found pathogenic microorganisms and milk chemical
changes. One of pathogens that cause mastitis is Staphylococcus species. This
study aimed to isolate and identify Staphylococcus species present in goat milk at
the "Sahabat Alam Sejati� Farm, Kalasan, Sleman, Yogyakarta.
A total of 40 samples of Ettawah cross breed goats milk were tested on
mastitis subclinical using reagents IPB-1. Thirty-seven positive samples of
subclinical mastitis cultured on Blood Agar media, incubated 37°C for 18 to 24
hours. The colonies were observed and grouped according to morphology and
Gram staining performed. Gram positive group were continued into catalase test.
The results of catalase positive were continued into coagulase test and the ability
of mannitol fermentation using Mannitol Salt Agar (MSA) media. Then for the
identification of Staphylococcus species, they were cultured on Voges Proskauer
(VP) media.
Result of the research found 27 isolates of Staphylococcus. The results of
identification obtained 9 isolates of Staphylococcus aureus (33,3%), 9 isolates of
Staphylococcus intermedius (33,3%), 5 isolates of Staphylococcus epidermidis
(18,5%), and 4 isolates of Staphylococcus hyicus (14,9%). |
