Summary:Mycobacterium tuberculosis is a gram-positive bacteri that causes tuberculosis disease. Tuberculosis is a zoonotic disease and the third cause of death after cardiovascular disease. Method and a proper diagnosis correct and quickly be required. This study aims to diagnose M.tuberculosis at levels concentration using PCR technique for reverens of tuberculosis detection. Materials used in this study is pure M.tuberculosis DNA from the Center for Veterinary Research. Initial phase is the DNA isolation. M.tuberculosis isolates were extracted, and then calculated the concentration of DNA quantitatively. DNA obtained was diluted to eight different concentrations of the 5000 pg/μl
