description | Tarsius sp. is a small nocturnal primate that found in Indonesia, which
decreased in population, and needs conservation attempt to preserve them. Species
of tarsius is very difficult to identify because of their morphology are nearly the
same and for its success of conservation, it needs encoding gene for them. The
main objective of this research is to study about the genetic diversity of NADH
Dehydrogenase Sub-unit 6 (ND6) encoding gene on Tarsius sp. DNA samples are
isolated from the thigh muscle of three Tarsius bancanus bancanus from
Lampung and a Tarsius dianae from Center of Celebes. DNA Samples than
amplificated with Polymerase Chain Reaction (PCR) technique used ND6F and
ND6R primer for T. b. bancanus samples and ND6F lama and ND6R primer for
T. dianae samples. Amplification process produces PCR product with 586 base
pair (bp) (T. b. bancanus) and 662 bp (T. dianae) which than sequenced.
Sequencing result then analyzed using MEGA 5.05 version program and
compared with some other Tarsius sp. species obtained from Genbank. Analysis
result showed that the genetic distance based on nucleotide sequence was 28,7%
and based on amino acid sequence was 27,1%. Construction of phylogeny tree
based on nucleotide and amino acid of ND6 gene with Neighbor-Joining method
could differentiate Tarsius bancanus bancanus and Tarsius dianae with Tarsius
sp. as comparing species. |
