Summary: | Isolation and structure elucidation of bioactive compounds and toxicity test and antibacterial test sponge Dysidea sp. have been done. The isolation was performed by extraction using a mixture of methanol:dichloromethane (1:1). Dichloromethane extract was than partitioned with ethyl acetate:water (3:2). Crude ethyl acetate extract was separated by column chromatography using a gradient elution system of n-hexane, dichloromethane, ethyl acetate and methanol. Crude ethyl acetate extract was also tested toxicity by the Brine Shrimp Lethality Test (BSLT) method and antibacterial test with agar diffusion method. Identification of bioactive compounds is done by MS and NMR spectroscopic methods. The result of toxicity test with BSLT method showed that Lethal Concentration, 50% (LC50) of crude ethyl acetat extract of Dysidea sp. was 219.78 ppm. The result of antibacterial test showed that the crude ethyl acetate extract of Dysidea sp. able to inhibit the growth of E. coli until the concentration of the test 25% and S. aureus until the concentration of the test 3.125%. MS and NMR spectroscopic methods analysis indicated that the compounds isolated from fraction 5 contains manzamenon D, thiofurodysinin acetate and β-sitosterol.
