Summary:Background: Identification is determining the identity of which is characteristic of something that distinguishes it from other different types. Many parameters can be used to identify one of them is the identification of ABO blood groups. The identification is not easy, it depends on the condition of the body such as the burning conditions. To determine the ABO blood group identified the body in a state of burning, use of molecular biology methods, namely PCR that has been proven high sensitivity. Objective: Detecting ABO blood group genotyping by burning the corpse bone organ samples using molecular biology methods which is namely PCR. Methods: Eight samples of bone (IV degree burns) were collected from the installation department of Forensic Medicine Dr. Sardjito Yogyakarta. Then, the DNA isolation and measurement of DNA concentration using a spectrophotometer. After this process, carried out by adding the composition of the PCR for reaction PCR. PCR product was then run in agarose gel for electrophoresis. Finally, the restriction is done by using an enzyme KpnI for 261 primary and 703 primary enzyme AluI for later read under UV light. Result: Shows the genotype and phenotype of the ABO blood group of eight bodies burnt bone samples which are detected, two samples with AA of ABO genotype and six samples with BB of ABO genotype with its phenotype A and B. Conclusion: It is concluded that molecular biology methods using PCR can detect blood group genotypes AA and BB with its phenotype A and B from eight samples of bone organ corpse burning conditions.
