Offspring from this crossbred called SimPO and LimPO which has advantages such as a large birth weight and rapid growth. The disadvantages of SimPO and LimPO cows are decreased of reproduction performance of Fenotipe 2 (F2) such as pregnancy rate being lower. Pregnancy stage will occur when oocyte had fertilized and had reached to embryos cleavage stage.The aims of this research were to determined in vitro rate of maturation, fertilization and cleavage embryos SimPO and LimPO oocytes. Ovaries were collected from local abbotoir grouped into PO (control), SimPO and LimPO. Cumulus-oocyte complexes were aspirated from ovaries with a 5ml syringe fitted with an 18 gauge needle. Cumulus-oocyte complexes quality A and B were used for this research. Cumulus-oocyte complexes were cultured in in vitro maturation medium droplets 100μl and covered by mineral oil at a 38.5°C, with 5% CO2 and 95% humidity for 24 hours. In vitro maturation rate determined by staining with 1% aceto orcein to define stage of oocyte after maturation by changes in chromosome configuration and the nuclear membrane stage, sucs as Germinal vesicle (GV), Germinal vesicle breakdown (GVBD), Metaphase I (MI), Anaphase I (AI), telophase I (IT) and Metaphase II (MII). Oocytes were fertilized using frozen semen of Simmental with concentration 5 x 106 cells / ml. In vitro fertilization rate to observed polymorphonuclear (PMN) formation 10 hours after insemination using 1% aceto orcein staining to fertilized oocytes. Fertilized oocytes were washed were transferred into culture and incubated at 38,5 ° C, with 5% CO2 and 95% humidity. Embryo cleavage rate were evaluated after 72 hours of incubation in tissue culture media 199. Research data were transformed to arcsin and analyzed by ANOVA one-way and followed by Fisher�s protected Least Significant Difference test. These results indicate that oocytes in vitro maturation rate reaching metaphase II (MII) in PO was significantly higher than Simpo and Limpo (P <0,05). In vitro fertilization rate of oocytes and cleavage embryos did not any significant differences between groups. Based on these results it could be concluded that SimPO and LimPO oocytes has in vitro maturation rate lower than PO. In vitro fertilization rate and embryo cleavage was not different between each group.
