antiinflammatory drugs (NSAIDs). The use of NSAIDs may cause some side
effects, and in some ways it may more secure to use medicinal herbs. Roselle
(Hibiscus sabdariffa) has potential effect as antiinflammatory agent. This study
aimed to know the effect of systemic administration of roselle ethanolic extract
toward COX-2 expression and neutrophils number in the inflammatory phase of
wound healing processes.
Thirty six of male white rats were given to injury on the buccal mucosa
by using punch biopsy. The animal models were divided into 3 groups randomly
into negative control (saline), positive control (ibuprofen 20 mg/kg), and
treatment (roselle 500 mg/kg). Rats were sacrificed on day 1st, 2nd , 3rd and day
4th prior to preparation of buccal mucosa tissues for paraffin blocks.
Immunohistochemical staining was performed to determine the expression of
COX-2, and hematoxylin eosin staining was done to determine the number of
neutrophils.
Two-way ANOVA of COX-2 expression and neutrophils number data
showed significant difference (p<0.05) in all groups. COX-2 expression and
neutrophils number was detected at lowest values on day 4th of roselle-treated
group. Clinical observation showed perfect wound closure in roselle-treated
group on day 4th. The results demonstrated that the ethanolic extract of roselle
could inhibit COX-2 expression and decrease the number of neutrophils. It might
be suggested that roselle ethanolic extract can be used as anti-inflammatory
agent of wound healing process. |
