description Caries is a bacterial infection which caused destruction of the tooth structure. Cleaning the cavity using cavity cleanser is one of the procedure to restore the tooth. This procedure aimed to kill the bacteria infect the dentinal tubules. Chlorhexidine digluconate (CHX) 2% is a common cavity cleanser use in daily dental practice. This agent has broad spectrum of antibacterial effect. The aim of this research was to study the effect of CHX 2% as a cavity cleanser to the continuity odontoblastic layer at the roof of the pulp chamber. Twenty four first molar teeth of 3-4 months old male Sprague dawley rats were used in this study. A cavity was made with 0,9 mm round diamond bur on the occlusal area of upper first molar. The rats were divided into two groups, each group consisted of 12 teeth. CHX 2% and aquadest were applied on the cavity surface of molar tooth of the rats in the treatment group and control group subsequently. The rats were sacrified at 1 hour, 3 days, 5 days, and 7 days after application. The specimens were decalcified using formic-acid HCl 5%, embedded in paraffin, and section serially. The sections were then stained using hematoxylin eosin. Observation were carried out using a light microscope with 400x magnification and assessed based on odontoblast layer continuity criteria. The assessment results of odontoblast layer were analyzed using Kruskal-Wallis test. The result of this research showed significant differences (p<0,05) between the control and treatment groups. In conclusion, application of CHX 2% as a cavity cleanser disrupt, the continuity of odontoblastic layer at the roof of pulp chamber
