Summary: | Protease enzyme is one of the most enzyme that classified into hydrolase
group, because this enzyme can catalyst the unbounded of peptide bond and have
been aplicated through many food or non-food industries. This enzyme can be
produce by fungus, one of them is Amylomyces rouxii. A. rouxii has a natural
habitat at sticky rice tape fermentation and can be isolated from ragi tape or
cassava tape . This study is aim to investigate the production condition of protease
enzyme from Amylomyces rouxii.
In this work, isolate was incubated in media from sticky rice, corn, or
soybean, then the protease activity is measured by solved protein content which is
produced by lowry method. Choosen media will be used to investigate the time,
the variation of basal salts and temperature effect on enzyme production. The A.
rouxii growth in sticky rice at 37oC for 3 days of incubation with variation basal
salts MgCl2, produce the highest activity which is 19.57 U/gram media compare
to that of anothers.
