| description | Purwoceng (Pimpinella pruatjan Molkenb.) is a medicinal plant that has commercial
properties as an aphrodisiac, diuretic and tonic. Great exploitation make P. pruatjan
becomes rare plants and endangered species as it has been reported by the Convention
on International Trading in Endangered Species of Wild Flora and Fauna (CITES).
The study was conducted to study at the genetic variability of P. pruatjan in Java
reviewed based on morphological and molecular characters using Inter Simple
Sequence Repeat (ISSR). Morphological and molecular characters of all accessions
were taken from four different in places such as the Dieng Plateau (Central Java),
Putri Mountain (West Java), Ranupane region (East Java) and Lawu Mountains
(Central Java). Morphological data retrieval was done by characterizing root, stems,
leaves, fruits and seeds. Molecular data was conducted by ISSR method and DNA
isolation was done with Qiagen DNeasy Plant Mini kit, while Polymerase Chain
Reaction (PCR) with 7 ISSR primers and electrophoresis. Dendrogram showed the
character of the vegetative formed two clusters with a similarity value of 90%.
Generative morphological characters formed two clusters with similarity value of
82.1%. Molecular character formed four clusters with a similarity value of 77.5%,
with an ISSR markers formed ecotypes based on sampling location. |
