Summary:More than 80% of chronic gastritis associated with Helicobacter infection pylori. Indonesia is known to have a high incidence rate of H.pylori infection, but the degree of gastritis severity and the risk of developing gastric cancer is very low. Various studies indicate that the Th-1 cytokines cause gastritis whereas Th-2 cytokines (including interleukin-10) have protective effect to the gaster. Interleukin- 10 (IL-10) is an anti-inflammatory cytokine that is considered to contribute a strong role in determining the type of endoscopic lesions of gastric mucosa with H.pylori infection. This study aims to determine differences in the expression of IL-10 is based on the severity of gastric mucosal lesions of chronic gastritis patients infected with H.pylori. This study is an observational study, the design of study is cross-sectional. Sample size will be determined by employing the rules of thumb, and it is established that the minimal sampel requirement should be 30 samples. And determined the amount of the next sample after test hypotheses put into the formula for the proportion of the population to 42 samples. Study population will be the gastric mucosal biopsy specimens of chronic gastritis patients (aged 18-80 years) who examination will be carried out in the department of Anatomical Pathology at Dr. Sardjito Hospital with positive H.pylori infection status since Januari 2009 until October 2013. Samples will be excluded if based on patient�s medical record data, the patient is known to have a history of malignancy, autoimmune disease, renal failure and chronic liver disease. Examination will be carried out in the department of Anatomical Pathology of Dr. Sardjito Hospital, Yogyakarta. Biopsy specimens that will be included in the data analysis are specimens with positive H.pylori infection status. Expression of interleukin-10 will be observed by using monoclonal antibodies that can recognize cytokines (interleukin-10) as well as a variety of cell markers used as primary antibodies and immunohistochemical staining performed by Avastin-biotinperoxidase method. Grading accuracy in this study will be assessed by kappa test (κ values is determined above 0,7).
