Summary: | Marker-assisted selection (MAS) is more accurate technique than conventional selection
because only individuals which have locus and gene target are selected. The objective of this
research was to determine the distribution of parental polymorphism and position of donor
segment on whole parental genome and also F1 verification associated with the quantiative trait
loci (QTL) for total spikelet number on chromosome 4 (qTSN4), day to heading on chromosome
8 (qDTH8) and Xa7 on chromosome 6. This research was conducted at Indonesian Center for
Agricultural Biotechnology and Genetic Resources Research and Development by February up
to September 2013. Three rice genotypes �Code, IR64-NILs-qTSN4[YP9] and IR64-NILsqDTH8[
YP1] and also their recurrent parent 'IR 64' were analyzed for polimorphism using 412
microsatellite markers and 1 STS marker, M5. Two 'IR 64' near isogenic lines (IR64-NILs) used
as parental for improving �Code� with higher yield, early maturity and also resistant to bacterial
leaf blight. The results show low percentage of polymorphism on two combination parent due to
three genotypes are used derived from 'IR 64'. Introgressed donor segment in �Code� speculate
that carrying genes related spikelet number per panicle besides qTSN4 so that �Code� has
spikelet number per panicle higher and significantly different than �IR 64� based on Dunnett test
W=5%. F1 verification using microsatellite markers RM17483, RM5556, RM6838 and RM20582
represent F1 'Code' � IR64-NILs-qTSN4[YP9] with number of plant 61-73 and also F1 'Code' �
IR64-NILs-qDTH8[YP1] with number of plant 86-100 can be used as backcross material with
'Code' to create BC1F1.
