Summary: | Background : Tuberculosis (TB) is a disease caused by
Mycobacterium tuberculosis. Tuberculosis is one of the
unslovable global health problem. The gold standard to diagnose
tuberculosis is bacterial culture. MGIT (Mycobacterium Growth
Indicator Tube)is one of liquid medium tuberculosis that
requires a relatively shorter time compared to culture in solid
medium. However, MGIT has a weakness where the contaminants are
easily to growth. One of the bacteria that is often as a
contaminant is Staphylococcus aureus. Anticipation for the
presence of these contaminant need to be done by using
antibiotics such as Piperacillin and Gentamicin.
Objective : To determine the effect of antibiotic Piperacillin,
Gentamicin or the mixture against Staphylococcus aureusin
culture of Mycobacterium tuberculosis by MGIT liquid medium.
Methods: This study used a culture of Mycobacterium tuberculosis
and Staphylococcus aureus (104 μg/ml)that are available in
Laboratory of Microbiology, Faculty of Medicine, University of
Gadjah Mada. The culture that�s performed in MGIT, is divided
into control group,which consist of control Mycobacterium
tuberculosis and control Staphylococcus aureus, as well as the
treatment groups, ie groups of Piperacillin, Gentamicin groups,
and mixed groups with MIC 2 μg/ml for Piperacillin and 8 μg/ml
for Gentamicin. Then, there�s further observation on the growth
of bacteria.
Results: There was a growth of Mycobacterium tuberculosis in
Piperasillin group without Staphylococcus aureus growth, that�s
demonstrated by the Mycobacterium tuberculosis-specific
turbidity and visually BTA on staining ZN, while there were no
growth any bacteria in gentamicin group and mixture group.
Conclusion: Piperacillin can be used to reduce the growth of
Staphylococcus aureus in MGIT. However, the antibiotics
gentamicin and Mixed (Piperacillin and Gentamicin) was unable to
inhibit the growth of Staphylococcus aureus in MGIT.
