description Daerah Istimewa Yogyakarta is one of the provinces in Java Island with several rivers flowing through it. One of them is Opak River with its dynamic ecosystem. Ahaetulla prasina and Ahaetulla mycterizans which are similar in morphology could be found in this ecosystem. This research aimed to find and add the morphologic, morphometric, and meristic characteristics that could be used to differentiate A. prasina and A. mycterizans, as well as to understand the characteristics� variations in the species in Opak River. Sampling was conducted day and night in 16 points along the upstream, middle, and downstream of Opak River, using a 500 m VES-transect for each point. Eighty-one characteristics in total were researched. Advanced identification and specimen verification was done in the Herpetology Laboratory, Research Center for Biology - Indonesian Institute of Sciences, Cibinong using three paratype specimens of A. prasina and five paratype specimens of A. mycterizans. Paratype specimen data were analyzed using MANOVA, followed by PCA for all specimens, and descriptive statistic analysis for Opak�s specimens. The results showed that there were 14 specimens of A. prasina and 11 specimens of A. mycterizans. Based on MANOVA results, there were 17 characters with significance rate less than 10%. Then, based on PCA results, there were 2 meristic characters that could differentiate A. prasina and A. mycterizans significantly. They were the amount of ventral scales and anterior dorsal scales. The type of morphologic variations of A. prasina and A. mycterizans specimens in Opak River was identic and showed no specific pattern. There were morphometric and meristic variations but they also showed no specific pattern. Based on the results, the conclusion were: the amount of ventral scales and anterior dorsal scales had a great influence on differentiate A. prasina and A. mycterizans. No other characteristics were found that could significantly differentiate A. prasina and A. mycterizans
