Summary: | Comb is an accessory organ on the head of chicken which is influenced by testosterone hormone and can be used as indicator of chicken�s fertility. Therefore this research was aimed to study the gene interaction of chicken comb inheritance from crossbreed between local chicken which has a pea comb and the backcross II which has a single comb and also to observe SOX5 gene polymorphism. The methods used in this research were taking care of parents and day old chick (DOC), blood collection, DNA isolation, Polymerase Chain Reaction (PCR), and Restriction Fragment Length Polymorphism (RFLP) using HindIII restriction enzyme. The results showed that crossbreed between local chicken and the backcross II have 2 different offspring. This crossbreed between pea comb (rrP-) and single comb (rrpp) showed the offspring have 100% pea comb. This result cannot to be determined the genotype of female parent with pea comb is rrPP or rrPp because lack of the amount of offspring. While genotype of DOC is heterozygote pea (rrPp). The result of SOX5 gene polymorphism showed that there was no polymorphism between pea and single comb. Conclusions of this research were the qualitative phenotype of offspring were different. Genotype of female parent which has pea comb cannot to be determined and there was no polymorphism of SOX5 between pea and single comb.
