In 2012 the project of �GAMA Ayam� has reached 3rd Backcrossed chicken generation (BC3). BC3 were placed to be a Parent Stock chicken capable to produces fertile eggs without broodiness. Prolactin (PRL) is a candidate for gene that intervence broodiness in chicken. The aim of this riset were to discover polimorphic region in PRL gene and analyze assosiation between PRL gene polymorphism with broodiness in GAMA Ayam. Seven pedigred Back Crossed chickens and two F1chickens were used. Chickens were intensively take care at University Farm (KP4) UGM. Broody behavior was obseved daily for six month. Blood samples were collectedfrom wing vein and genomic DNA was isolated following protocol Roche®. Polymerase Chain Reaction (PCR) method was used to amplify 192 bp target PRL gene. Polymorphic trait showed with Single Stranded Comformation Poymorphism (SSCP) method. Subsequentgel was stained with etidium bromide to visualize banding patterns of the DNA fragment. Result of SSCP showed that polymorphism in PRL gene didn�t detected. Furthermore broody behavior is regulated by many gene (polygenic).
