Description
Summary:The production of embrio in vitro needs good quality oocytes. Factor that affected the quality of oocytes are Body Condition Score (BCS) and collecting method. The purpose of this study was to determined the effect of BCS to quality of oocytes morphology collected in vitro. Oocytes were collected from slaughter beefs ovary at Giwangan slaughter house, Yogyakarta. The ovaries differentiated by the level of BCS between 2 - <3 and 3 - <4 on a scale of 1-5. Ovary cleaned and the mesovarium washed twice with 0.9 % NaCl physiological solution which has been coupled with Penicyllin - Streptomycin antibiotics and put into a flask containing with same solution at a temperature of 350-370 C and then brought to the laboratory. Oocyte collected by aspiration using a 10 ml syringe filled with a solution of Phosphate Buffer Saline (PBS) as much as 0.5-1 ml with 18 gauge needle. Follicular fluid were obtained inserted tube and added 5 ml PBS, precipitated 15 minutes. The supernatant was discarded and the remainder was poured in a petri dish and then viewed under a microscope to clasified based on morphologic (A, B, C, D). The obtained results were analyzed by T-test. The results of the analysis of the quality of oocytes obtained A, B, C, D and A + B respectively (P = 0.804), (P = 0.596), (P = 0.272), (P = 1.000), (P = 0.838) showed no significant difference. The conclusion of this study is that the BCS does not affect the quality of the morphology of oocytes collected in vitro (P> 0.05).
