Summary: | Isolation, structure identification, and toxicity test of bioactive compounds from sponge Clathria sp. collected in Barrang Lompo Sea, South Sulawesi had been carried out. Isolation was performed by maceration with a mixture of methanol:dichloromethane (3:2 v/v). The dichloromethane extract was then concentrated and partitioned in ethyl acetate:water (1:1 v/v). Ethyl acetate fraction was separated and evaporated to obtain crude extract. The crude extract was separated by column chromatography with increasing polarity of the eluents from n-hexane, chloroform, dichloromethane, ethyl acetate to methanol. The toxicities of crude extract and fractions of column chromatography were tested using Brine Shrimp Lethality Test (BSLT) method. Fractions containing active compounds were analysed by Liquid Chromatography-Mass Spectrometry (LC-MS).
Crude extract obtained from the extraction is 0.613 g and the value of Lethal Concentration 50% (LC50) is 78.60 ppm. Nine fractions were collected from column chromatography separation. Fraction III, V, and VII showed high toxicity at single test concentration of 83.9 ppm. Toxicity test using BSLT method revealed the fraction III, V, and VII have LC50 values of 188.34
