Summary:The main objective of conducting this research is to better understand the impact of breed towards the quality of oocyte morphology that has been collected through in vitro, and also determine one of factors that support the efficiency of oocyte collection method for in vitro fertillization.(IVF) The oocyte was taken from the ovarium of cattle which taken from abbatoir (RPH) in Giwangan, Yogyakarta. The ovarium is classified based on the breed which include simPO, limPO, and PO. It was cleaned from mesovarium and its fat, then it was being washed using 0.9% of physiological NaCL which has been added by Penicyllin-Streptomycin antibiotics for twice and then it put into a flask containing the same solution at a temperature of 35 º - 37 º C to be taken straight to laboratorium. Oocyte collection is done by aspiration using a 10 ml syringe with a 18 gauge needle that has been filled in Phosphate Buffer Saline solution (PBS) of 0.5-1 ml. The liquid obtained was put in a tube and added by 5ml of PBS, then precipitated for 15 minutes. The supernatant was then discarded and the remainder was poured into a petri dish to be observed using a microscope to be classified based on its morphology. The obtained results were anlyzed by Anova method (P<0,05). The result of the anlysis of the quality of oocytes obtained A, B, C, and D respectively (P=0,60), (P=0,71), (P=0,314), and (P=0,079) showed no significant difference. The conclusion of this study is that the breed does not affect the quality of the morphology of oocytes collected in vitro.
