Summary:Streptococcus suis is a bacterial that cause arthritis, meningitis, bronchopneumonia and septicemia in pigs and kown as a zoonotic disease. In this research were used 67 tonsil swab samples from pigs in the various area in Papua. Identification of Streptococcus suis were performed with Gram staining, 6,5% NaCl selective media, Voges Proskauer (VP) and biochemical test. The results showed that 27 Streptococcus suis isolates were Gram-positive, negative reaction on the 6,5% NaCl and VP, able to ferment the glucose and trehalose (40,3%) as well as arabinose (25,37%), raffinose (38,85%) and sorbitol (31,34%). Characterization of Streptococcus suis isolates were performed based on the nature of growth in sheep blood agar plates, broth media and Soft Agar (SA). Streptococcus suis grew with small colonies, partially transparent and capable to hemolyse of blood agar plates revealed an α hemolysis for 27 isolates (40,3%). Nineteen isolates (28,36%) grew cloudy in liquid media and diffuse in the SA media, 8 isolates (11,94%) grew with the clear supernatant in broth media and compact on the SA media.
