Summary: | Trimeresurus puniceus is a viper that can be found on Java and southern
Sumatra. Snake envenomation an important and often neglected source of human
suffering especially in developing country including Indonesia. Currently there is
no monovalent anti-venom and effect treatment for patient bitten by Trimeresurus
puniceus. The objective of this research is to study the effect of Trimeresurus
puniceus venom on the morphology and the aggregation and hemolysis of cow�s
red blood cell.
The materials used in this research are the venom of Trimeresurus
punicues and cow erythrocyte. The research was carried out by diluting the venom
of Trimeresusrus puniceus with phosphate buffer saline (PBS) into concentrations
of 10%, 1% and 0.1% and each concentration was mixed with 100% cow
erythrocyte in 1:1 ratio, then rest it for 5 minutes. The red blood cell morphology
was examined with a microscope and the pattern of aggregation and henolysis was
estimated with red blood cell number using a hemocytometer.
All the red blood cell morphology undergoes multiple changes including
changing of biconcave shape to spherocytes, aniscocytosis, and echinocytes. The
venom of Trimeresurus puniceus cause aggregation and hemolysis in red blood
cell in all the 3 concentration that we tested, with roughly 73% aggregation and
hemolysis in venom with 10% concentration. It was concluded that the venom of
flat nose pit viper (Trimeresurus puniceus) has high hemolytic action and the
effect is higher with the increase of venom concentration.
