Safety evaluation needs to be done by carrying out acute toxicity tests to establish the acute toxicity potency, find out the arising toxic symptoms and observe organ histopathology caused by acute oral administration of earthworm (Lumbricus rubellus) powder. Acute toxicity test conducted on Swiss strain male mice based on OECD Guideline 423 with an initial dose of2000 mg/kg body weightfrom some ranks that can be used, those are 5, 50, 300 and 2000 mg / kg body weight. The analysis includes the acute toxicity potency (LD50), observation of toxic symptoms and organ histopathology. Test substance does not cause toxic symptoms or death that can not be tolerated up to day 14 after oral administration. Based on the OECD Guideline 423, the test substanceclassified in category 5 (not classified) with LD50cut-off > 5000 mg/kg. Histopathological observations showed that the test substancedoesn�t have toxic effect on the lungs, stomach, spleen, heart, intestine, testis and kidney in male mice. However, fatty degeneration was found occuring in 33,3% mice dose group 5000 mg/kg body weight.
