description Kaffir lime (Citrus hystrix D.C) is an herbal plant which widely spread in Indonesia. During this time, the use of plants is still limited as food ingredients and aromatherapy oils. Leaves of C. hystrix contain flavonoids, alkaloids, saponins, polyphenols, terpenoids, tannins, and essential oils that are cytotoxic to leuchemic T-cell and cervical cancer cell line. Objectives of this study were to determine cytotoxicity and apoptotic induction of C. hystrix leaves extract on T47D cells and Vero cells. Methods that used in study were maceration using three solvents (ethyl acetate, chloroform, and ethanol 96%), cytotoxic assay by MTT assay, apoptotic assay using double staining (ethidium bromide-acrydine orange). The results showed that cytotoxicity assay of chloroform extract (IC50= 128,89 μg/mL ) have the highest cytotoxic activity on T47D cells compare to ethyl acetate extract (IC50= 283,12 μg/mL) and ethanolic extract (IC50= 444 μg/mL). Whereas in Vero cells three extract were not reached IC50 hence these extract had no cytotoxic effect in non-cancer cells. Furthermore apoptotic rate of extract chloroform, ethyl acetat, and ethanol 96% were (51,45%), (31,47%), (36.24%) respectively. Chloroform extract kaffir lime leaves most potential to induce apoptosis of T47D cells followed by ethyl acetat and ethanol.
