description | Background : Tuberculosis (TB) is a disease caused by
Mycobacterium tuberculosis. It is difficult to control the
transmission of this disease and the treatment cost is high and
it takes a long time to recover. These facts make TB one of the
major health problems in the world. The gold standard to
diagnose tuberculosis is bacterial culture. MGIT (Mycobacterium
Growth Indicator Tube) is one of the liquid culture medium which
could be used to detect TB in a relatively shorter time compared
to solid culture medium. However, the weakness of MGIT is it�s
high incidence of contamination. One of the possible bacteria
which could contaminate the medium is Pseudomonas sp .. In order
to prevent the growth of the contaminants, it is necessary to
use antibiotics such as Piperacillin and Gentamicin.
Objective : To determine the effect of antibiotics
Gentamicin, Piperasilin or the mixtures against Pseudomonas sp.
in Mycobacterium tuberculosis cultures using MGIT liquid medium.
Methods : This study used cultures of Mycobacterium
tuberculosis and Pseudomonas sp. (104 μg/ml) which were provided
by the Laboratory of Microbiology, Faculty of Medicine,
University of Gadjah Mada. The culture are performed in MGIT
culture medium and are divided into 5 groups: control group,
which include control of Mycobacterium tuberculosis and control
of Pseudomonas sp., As well as the treatment group, i.e.
Gentamicin group, Piperacillin group, and the Mixed group with
MIC 8 μg/ml for Gentamicin and 2 μg/ml for Piperacillin.
Afterwards, further observation on the growth of bacteria was
done.
Results : The growth of Mycobacterium tuberculosis and
Pseudomonas sp. in Piperacillin group was detected. It was
indicated by positive results from the microscopic examination
and culture. Whereas there was no growth of any bacteria in
Gentamicin group and the mixture group.
Conclusion : Both antibiotics, Gentamicin and Piperacillin
have no potential to reduce the growth of Pseudomonas sp. in
MGIT |
