description Analgesic and anti -inflammatory had been found and having potential for therapy but most of them have side effects. Breadfruit (Artocarpus altilis) is a plant which had not been used maximally yet for medication. The purposed of this research to determine analgesic and anti -inflammatory effect of the aethyl acetate extract from the breadfruit leaves (E ADS) and also determine the COX-2 expression and it�s selectivity . The analgesic potency was done by acetic acid induced to mice. Potency of antiinflammatory was done by carragenan induced in the soles of their paw. Immunohistochemistry test was done by observed the soles of mice�s paw carragenan reduced to express the COX-2 enzyme. The selectivity of the EADS extract to COX-1 and COX-2 inhibition was done by colorimetric COX (Ovine) with Inhibitor Screening Assay Kit (Cayman). EADS extract doses of 250, 500 and 1000 mg/kgBW had analgesic activity of (22.92 ± 5.02), (38.54 ± 3.61) and (63.22 ± 2.91)%. Analgesic activity of extracts EADS still had smaller than aspirin (55 mg / kg) that had analgesic activity of (86.90 ± 4.49)%. EADS extract doses of 250, 500 and 1000 mg/kgBW of antiinflammatory activity (18.04 ± 5.01), (28.12 ± 9.59) and 47.08 ± 7.46)%. Antiinflammatory activity of EADS extract was still smaller than Indomethacin (37.2 mg/kgBW) that had antiinflammatory activity (56.37 ± 2.36)% and diclofenac sodium (19.5 mg / kg) that had antiinflammatory activity (52.52 ± 9.34)%. EADS extract doses of 250, 500 and 1000 mg/kgBW, was able to decrease the expression of COX-2 (21.62 ± 3.59), (36.33 ± 2.26) and (45.77 ± 4.71) % respectively . The positif control, Indomethacin (37.5 mg/kgBW) and Diclofenac sodium (19.5 mg/kgBW) had activity more stronger with decreased respectively (66.08 ± 2.81)% and (73.76 ± 0.73)%. IC50 value of the extract EADS inhibition of COX-1 and COX-2 was 23.45 µg/ml and 3.17 µg/ml respectively. EADS extract had analgesic and antiinflammatory activity, inhibiting the expression of COX-2 and inhibit the activity of COX-2 more selective than COX-1. This research can be used for developing of EADS extract as a source of compounds with analgesic and antiinflammatory activity.
