description | Melon (Cucumis melo L.) is one of susceptible horticulture to disease infections, including viral infections and the most harmful viruses nowadays is Begomovirus. Begomovirus infection causes plants become chlorotic, curly, and stunted. It causes huge economic losses for farmers. Cultivar Melodi Gama 3 (MG3) becames candidate resistance crops for Begomovirus�s infections. Development of Sequence Characterized Amplified Region (SCAR) from Random Amplified Polymophisme DNA (RAPD) was done in this research by DNA sequencing and Polymerase Chain Reaction (PCR) methods. In addition, detection DNA particles of Begomovirus on melon cultivar MG3 has been carried out using Coat Protein (CP) gene primers. The results showed that the SCAR markers linkage to resistance gene of melon against Begomovirus successfully developed from RAPD. Further results showed Squash leaf curl Philippines virus is one of the Begomovirus strain infected melon MG3. Whereas chi-square test determined that the observed data appropriated for the expectations data at a low level test (0.01), hence application of SCAR markers on a larger population is needed to determine the inheritance pattern of resistance characteristic of cultivar MG3 against Begomovirus infection. The final results of this study concluded that the resistance character of cultivar MG3 expectedly controlled two types of independent genes. |
