The research was subjected to detect local isolate Toxoplasma gondii antigen by Sandwich ELISA with TL106 specific monoclonal antibody toward local isolate T. gondii membrane antigen for the earlier detection and the development of toxoplasm diagnosis. This research was compared to in vivo cultivation of T. gondii, injection of T. gondii on experimental animal, antibody purification, sample colection, and detection of serum antigen in infected animals. Seventy Swiss webster female mice in the age of 2 months were alotted to 7 groups and 10 of each. The six groups of it were injected with 1x106,1x105,1x104,1x103, 1x102 and 1x101 tachyzoite respectively, and another was injected with physiologic sodium saline (as control group). Sera were colected two days after treatment. The obtained sera were tested by Sandwich ELISA with TL106 specific monoclonal antibody which optimised before. The sensitivity of the test were paraleled with Toxo-ISAGA aglutination test. The result showed that the Sandwich ELISA with TL 106 specific monoclonal antibody toward local isolate T.gondii membrane antigen had sensitivity value 90 %, the positif assesment value 94,7 %. The research was concluded that the TL 106 specific monoclonal antibody toward local isolate T. gondii membrane antigen abled to detect T. gondii antigen earlier and the T. gondii antigen could be detected before IgM produced in toxoplasmosis.
