Description
Summary:ABSTRACT Daryono, B.S. 2006. Resistance to cucurbit viruses in several genotypes of melon (Cucumis melo L.). Berkala nmiah Biologi 5 (1): 1 -12. Virus diseases are important problem for production of melon in the world. Among the diseases, Cucumber mosaic virus (CMV), Zucchini yellow mosaic virus (ZYMV), Watermelon mosaic virus (WMV), and watermelon strain of Papaya ringspot virus (PRSV-W) have been reported occurring in field-grown cucurbits in Indonesia. To elucidate genetic source of resistant melons, 60 melons collected from 19 Asian countries were screened for resistance to CMV, PRSV, Cucumber green mottle mosaic virus (CGMMV), and Kyuri green mottle mosaic virus (KGMMV). Levels of resistance to virus accumulation in melon leaf tissue were evaluated using a combination of symptom observation, Double Antibody Sandwich-Enzyme Linked Immunosorbent Assay (DAS-ELISA) and Reverse Transcriptase-Polymerase Chain Reaction (RT-PCR) analysis. Melon cultivars responded differently to the viruses. Melons culti-var Yamatouri, Mawatauri, Kohimeuri, PI 161375, and PI 371795 showed to be completely resistance to CMV, while resistance to PRSV-P was found in cultivars Yamatouri, Mawatauri, PI 161375, PI 414723, and PI 371795. Moreover, cultivars Mawatauri, Kohimeuri, PI 161375, and PI 371795 showed resistance to KGMMV. In contrast, all melons tested showed susceptible to CGMMV. Keywords: Cucumis melo L., cucurbit viruses, DAS-ELISA, RT-PCR.
