The aim of this study were to detect circulating antigen of Toxoplasma gondii using 129 monoclonal antibody to membrane antigen of local isolated Toxoplasma gondii as an early detection for toxoplasmosis and to know the prevalence of toxoplasmosis of slaughterhouse in Yogyakarta district.
In this study the specimens from cows (80 specimens), sheep (80 specimens), and pregnant women (20 specimens) were used respectively. The samples were tested for toxoplasmosis using ELISA Sandwich Test. Checkerboard was done before ELISA Sandwich Test to determine antibody dilution and antibody choice. Sample from pregnant women which has been tested by commercial serology, that is Toxo ISAGA and Toxo Screen DA, were then tested by ELISA Sandwich Test. The comparison of Toxo ISAGA, Toxo Screen DA, and ELISA Sandwich results was used to determine sensitivity test.
The result showed that the use of 129 monoclonal antibody to membrane antigen of local isolated Toxoplasma gondii had 88.8% sensitivity. The prevalence of toxoplasmosis was 27.5% in cow and 76.25% in sheep. It could be summarized that 129 monoclonal antibody of local isolated Toxoplasma gondii might be used to detect circulating antigen of toxoplasmosis cases in the early stage
