description Regeneration of peanut plant through somatic embryogenesis was the most effective way for plant propagation. Research on peanut somatic embryogenesis so far was quite limited, so that the present one was done with objective to determine the best combination of 2,4-D and sucrose was the most effective in promoting peanut somatic embryogenesis, and to study the protein configuration in early phase of explant development as affected by 2,4-D and sucrose. A 2x3 factorial experiment argumented with a control treatment was initiated in completely randomized design. The first factor, 2,4-D was tested in two concentrations: 10 and 20 mg/1. The second one was sucrose concentration which ranged from 20,30 to 40 g/1. Data collected were subjected to analysis of variance followed by mean separation based on Duncan's Multiple Range Test. Protein configuration was determined using SDS-PAGE 10%. The result showed that application of 20 mg/1 2,4-D and 40 g/1 sucrose produced somatic embryo at considerable number in relatively short time. There were differences in protein configuration in the early phase of explant development, particularly at 2 and 4 days after culture initiation, but similar protein configuration was observed in the later periods.
